Early studies demonstrated the involvement of ubiquitination of the Notch intracellular domain for rapid turnover of the transcriptional complex at Notch target genes. It was shown that this ubiquitination was promoted by the co-activator Mastermind like 1 (MAML1). MAML1 also contains numerous lysine residues that may also be ubiquitinated and necessary for protein regulation. In this study, we show that over-expressed MAML1 is ubiquitinated and identify eight conserved lysine residues which are required for ubiquitination. We also show that p300 stimulates ubiquitination and that Notch inhibits ubiquitination. Furthermore, we show that a mutant MAML1 that has decreased ubiquitination shows increased output from a HES1 reporter gene assay. Therefore, we speculate that ubiquitination of MAML1 might be a mechanism to maintain low levels of the protein until needed for transcriptional activation. In summary, this study identifies that MAML1 is ubiquitinated in the absence of Notch signaling to maintain low levels of MAML1 in the cell. Our data supports the notion that a precise and tight regulation of the Notch pathway is required for this signaling pathway.

The Notch signaling pathway is an evolutionarily conserved system found in all metazoans and it plays an important role in developmental and disease processes by influencing proliferation, differentiation, self-renewal and apoptosis [1, 2]. In mammals, there are four Notch receptors. The Notch receptor is a protease-activated transcription factor located on the cell membrane. Interaction with a ligand results in cleavage releasing the intracellular domain (NICD). The NICD enters the nucleus and forms a complex with CSL (CBF1, Suppressor of Hairless, and Lag-1) enhancer-binding proteins and MAML1 to activate target genes. The ternary complex (CSL, NICD, and MAML1) is believed essential for activation of target genes (Fig 1). MAML1 was initially identified as a protein involved in the Notch pathway in D. The purpose of this study was to determine how drainage impacts carbon densities and recent rates (past 50 years) of vertical accretion and carbon accumulation in southeastern forested peatlands. We compared these parameters in drained maple-gum (MAPL), Atlantic white cedar (CDR), and pocosin (POC) communities in the Great Dismal Swamp National Wildlife Refuge (GDS) of Virginia/North Carolina and in an intact (undrained) CDR swamp in the Alligator River National Wildlife Refuge (AR) of North Carolina. Peat cores were analyzed for bulk density, percent organic carbon, and 137Cs and 210Pb. An uncertainty analysis of both 137Cs and 210Pb approaches was used to constrain error at least partially related to mobility of both radioisotopes. GDS peats had lower porosities (89.6% (SD = 1.71) versus 95.3% (0.18)) and higher carbon densities (0.082 (0.021) versus 0.037 (0.009) g C cm-3) than AR. Vertical accretion rates (0.10-0.56 cm yr-1) were used to estimate a time period of 84-362 years for reestablishment of peat lost during the 2011 Lateral West fire at the GDS. Carbon accumulation rates ranged from 51 to 389 g C m-2 yr-1 for all sites. In the drained (GDS) versus intact (AR) CDR sites, carbon accumulation rates were similar with 137Cs (87GDS versus 92AR g C m-2 yr-1) and somewhat less at the GDS than AR as determined with 210Pb (111GDS versus 159AR g C m-2 yr-1). Heightened productivity and high polyphenol content of peat may be responsible for similar rates of carbon accumulation in both drained and intact CDR peatlands.


Background The ParaHox genes are thought to be major players in patterning the gut of several bilaterian taxa. Though this is a fundamental role that these transcription factors play, their activities are not limited to the endoderm and extend to both ectodermal and mesodermal tissues. Three genes compose the ParaHox group: Gsx, Xlox and Cdx. In some taxa (mostly chordates but to some degree also in protostomes) the three genes are arranged into a genomic cluster, in a similar fashion to what has been shown for the better-known Hox genes. Sea urchins possess the full complement of ParaHox genes but they are all dispersed throughout the genome, an arrangement that, perhaps, represented the primitive condition for all echinoderms. In order to understand the evolutionary history of this group of genes we cloned and characterized all ParaHox genes, studied their expression patterns and identified their genomic loci in a member of an earlier branching group of echinoderms, the asteroid Patiria miniata. Results We identified the three ParaHox orthologs in the genome of P. miniata. While one of them, PmGsx is provided as maternal message, with no zygotic activation afterwards, the other two, PmLox and PmCdx are expressed during embryogenesis, within restricted domains of both endoderm and ectoderm. Screening of a Patiria bacterial artificial chromosome (BAC) library led to the identification of a clone containing the three genes. The transcriptional directions of PmGsx and PmLox are opposed to that of the PmCdx gene within the cluster. Conclusions The identification of P. miniata ParaHox genes has revealed the fact that these genes are clustered in the genome, in contrast to what has been reported for echinoids. Since the presence of an intact cluster, or at least a partial cluster, has been reported in chordates and polychaetes respectively, it becomes clear that within echinoderms, sea urchins have modified the original bilaterian arrangement.
