The formation of thrombi in the tail artery of mice receiving intravenous injection of Escherichia coli endotoxin was studied. Thrombus formation was more marked in endotoxin-treated than in saline-treated animals. Saline-treated mice showed a strong thrombocytopaenia after 24 hr, but endotoxin-treated mice showed only a moderate thrombocytopaenia. Thrombus formation and platelet consumption in the arterial thrombi were similar in mice receiving E. coli endotoxin and mice treated with adenosine diphosphate or arachidonic acid, respectively. The thrombotic response in mice treated with the synthetic prostaglandin synthetase inhibitor indomethacin (5 mg/kg) was attenuated. Platelets from endotoxin-treated mice were stimulated to produce eicosanoids, as measured by aggregation and release of arachidonate metabolites, after a short exposure to indomethacin.
